Multimodal regulation of encystation in Giardia duodenalis revealed by deep proteomics

چکیده

Cyst formation in the parasitic protist Giardia duodenalis is critical to its transmission. Existing proteomic data quantifies only 17% of coding genes transcribed during encystation and does not cover complete process from trophozoite mature cyst. Using high-resolution mass spectrometry, we have quantified changes across compared this with published transcriptomic data. We reproducibly identified 3863 (64.5% proteins) 3382 proteins (56.5% over standard growth (TY), low-bile priming (LB), 16 h into (EC), at cyst maturation (C). This work provides first known expanded observation level triples coverage previous proteomes. Encystation commitment in Giardia duodenalis: a long and winding road.

Cholesterol and bile salts are relevant modulators of Giardia encystation. Although several molecules within signaling cascades have been identified, and changes in their expression observed during giardial encystation, their underlying interactions leading to expression of cyst wall markers (CWPs and precursors of the GalNAc homopolymer) are not well defined. Recent experimental data and the c...

The Proteome Landscape of Giardia lamblia Encystation

Giardia lamblia is an intestinal protozoan parasite required to survive in the environment in order to be transmitted to a new host. To ensure parasite survival, flagellated trophozoites colonizing the small intestine differentiate into non-motile environmentally-resistant cysts which are then shed in the environment. This cell differentiation process called encystation is characterized by sign...

Cultivation of Giardia duodenalis in Mongolian gerbils.

The Mongolian gerbil (Meriones unguiculatus) is susceptible to infection with Giardia duodenalis trophozoites. Each animal was orally infected with 0.5 ml Diamond's TYIS-33 culture medium containing 10(6) trophozoites. Cysts were then collected and concentrated by sucrose gradient centrifugation. G. duodenalis cysts were first observed in feces on day 5 post-infection. The characteristic of G. ...

Genotyping and Descriptive Proteomics of a Potential Zoonotic Canine Strain of Giardia duodenalis, Infective to Mice

The zoonotic potential of giardiasis, as proposed by WHO since the late 70's, has been largely confirmed in this century. The genetic assemblages A and B of Giardia duodenalis are frequently isolated from human and canine hosts. Most of the assemblage A strains are not infective to adult mice, which can limit the range of studies regarding to biology of G. duodenalis, including virulence factor...

Comparative evaluation of Giardia duodenalis sequence data.

A review of the Giardia duodenalis sequences currently available on the GenBank database was completed to compare the different genotyping loci (small subunit ribosomal DNA, glutamate dehydrogenase, triose-phosphate isomerase and beta giardin) for their ability to discern assemblage and subassemblage groups and infer phylogenetic relationships. In total, 405 Giardia duodenalis sequences were so...
